Mechanism Of Action Of Tirzepatide

  • Tirzepatide acts on both GLP-1 and GIP receptors as a dual agonist.
  • It improves insulin sensitivity, decreases glucagon secretion, encourages satiety, and intensifies the insulin response.
  • Additionally, tirzepatide was found to slow down gastric emptying, lower fasting, and postprandial glucose levels, reduce food intake, and total body weight.

 

Therapeutic Indications Of Mounjaro Pre-Filled Pen

  • In people with type 2 diabetes mellitus, Mounjaro is recommended as an addition to diet and exercise to enhance glycemic control.

 

Method Of Administration Of Mounjaro Pre-Filled Pen

  • Always take this medicine exactly as your doctor or pharmacist has told you.
  • Check with your doctor or pharmacist if you are not sure.
  • The recommended starting dose is 2.5 mg given subcutaneously once weekly.
  • Your doctor may increase the dose to a once-weekly injection of 5 mg subcutaneously after 4 weeks.
  • After taking the current dosage for at least 4 weeks, your doctor may increase the current dose by 2.5 mg if more glycemic control is required.
  • The highest dose is 15 mg administered subcutaneously once per week.
  • In all doses, administer once a week, with or without meals, at any time of the day.
  • While using mounjaro, consider the following precautions:
  1. Before you use the pen for the first time, ask your doctor or pharmacist to show you how to use the pen.
  2. You can use this medication at any time of the day, with or without food and drink.
  3. Use this medication at about the same time 
  4. The best places to inject are the front of your waist (abdomen), the front of your thighs, or your upper arm.
  5. Do not inject into a vein or muscle.
  6. Do not mix this medication up with other medicines that you inject (e.g. insulins).
  7. Change (rotate) your injection site with each injection. Do not use the same site for each injection
  8. Doses may be changed in special conditions so always follow your doctor's or pharmacist's instructions.
  9. If you take more than you should, talk to a doctor or go to the hospital straight away.
  10. If you forget to take a dose, take it as soon as you remember it unless it is nearly time for your next dose.
  11. Do not take a double dose (two doses at the same time) to make up for the forgotten dose.

 

 

Undesirable Side Effects Of Mounjaro Pre-Filled Pen

  • Although the following side effects are not common in everyone, some patients may complain from:
  1. Nausea, diarrhea or constipation, and vomiting
  2. Decreased appetite
  3. Dyspepsia and abdominal pain.

 

Pregnancy & Breastfeeding Precautions While Taking Mounjaro Pre-Filled Pen

  • If you are pregnant or breastfeeding, think you may be pregnant, or are planning to have a baby, ask your doctor or pharmacist for advice before taking this medicine.
  • Your doctor will discuss the possible risks and benefits of using this medicine during pregnancy and breast-feeding

 

Non-Pharmacological Treatments & Advice For Diabetic Patients

  • All carbohydrates affect blood glucose levels so it’s important to know which foods contain carbohydrates. Choose the healthier foods that contain carbs and be aware of your portion sizes.
  • Eat less salt. Eating lots of salt can increase your risk of high blood pressure, which in turn increases the risk of heart diseases and stroke. And when you have diabetes, you’re already more at risk of all of these conditions.
  • Eat more fruit and vegetables. This can help you get the vitamins, minerals, and fiber your body needs every day to help keep you healthy.
  • You can always try low or zero-calorie sweeteners (also known as artificial sweeteners) to help you cut down sugar for example Steviana Sweetener or Tropicana Slim Sweetener
  • Don’t forget to keep moving. Being more physically active goes hand in hand with eating healthier.
  • Remember to measure your blood glucose level continuously by using home monitor.

  • Generally, weight loss occurs when energy intake is lower than energy expenditure.
  • People with obesity need to get at least 150 minutes a week of moderate-intensity physical activity to prevent further weight gain or to maintain the loss of a modest amount of weight.
  • You probably will need to gradually increase the amount you exercise as your endurance and fitness improve.

Warnings & Special Precautions While Taking Mounjaro Pre-Filled Pen

  • Before using Mounjaro, tell your healthcare provider if you have any other medical conditions, including if you:
  1. Personal or family history of medullary thyroid carcinoma.
  2. Known serious hypersensitivity to tirzepatide or any of the
    excipients.
  • Tell your doctor or pharmacist if you are taking, have recently taken, or might take any other medications including over-the-counter non-prescription medicines, prescription medicines, herbal supplements, vitamins, and supplements.

 

Storage Conditions Of Mounjaro Pre-Filled Pen

  • MOUNJARO is a clear, colorless to slightly yellow solution.
  • store in a refrigerator (2°c – 8°c).
  • Once needed, a single-dose pen can be kept unrefrigerated for up to 21 days at temperatures not higher than 30°C.
  • Keep this medicine out of the sight and reach of children.
  • Store in the original package to protect it from moisture.
  • Do not use this medicine after the expiry date which is stated on the box/blisters/label. The expiry date refers to the last day of the month.

 

Normal Ranges

  • Random blood sugar test: A blood sample will be taken at a random time. Regardless of when you last ate, a blood sugar level of 200 milligrams per deciliter (mg/dL) or higher suggests diabetes. A blood sugar level of less than 140 mg/dL is normal. A reading between 140 and 199 mg/dL indicates prediabetes
  • Fasting blood sugar test. A blood sample will be taken after an overnight fast. A fasting blood sugar level of less than 100 mg/dl is normal. A fasting blood sugar level from 100 to 125 mg/dL is considered prediabetes. If it's 126 mg/dL or higher on two separate tests, you may have diabetes.
  • Oral glucose tolerance test: For this test, you fast overnight, and the fasting blood sugar level is measured. Then you drink a sugary liquid, and blood sugar levels are tested periodically for the next two hours.
